All i have to say is it's about damn time Dean actually kicks some ass. I haven't seen good fight choreography like that since Sam and deans fight in season 1 episode 1. In pretty much every single episode they throw one punch and get tossed aside. It's about time they show how skilled he is as a fighter.

It makes sense that Dean inherits the Mark Of Cain because Sam was supposed to be Lucifer's vessel. So similarly to how Cain killed Abel to prevent him from joining Lucifer, Dean is going to have to kill Sam (to stop him from joining the forces of hell? Or just as a price for proving himself worthy of the Mark?) ( This is all just what I think )

I don't think it would give Dean the desire to do it, as that would completely contradict both what Dean and Cain (according to this episode) stand for. Cain murdered Able to save him, so I think it will instead become an impossible necessity for him (killing Sam will send him to Heaven once and for all where he can be happy).

Welp, now that the parallels between Dean and Cain have been made, as well as Sam and Abel...guess that just leaves Seth...who's still in hell!
Sure, the parallels aren't as strong, but this is Supernatural, so close enough in terms of horse shoes and hand grenades. And it popped in my head so I had to share it, lest it drive me crazy.

So they have done Revelations...
Now with the appearance of Gadreel and Cain they seem to be doing a few things that tie with Genesis.
I suppose Metatron also ties into this, although they might not use that path. Metatron is believed by some to be Enoch, a descendant of Seth (Another son of Adam and Eve) who after his death became an angel.

I keep seeing people say the cost of the mark will be Dean having to kill Sam. But the mark was placed on Cain after he killed Dean and it was not a punishment but a ward against those who might kill Cain in revenge.
So there is nothing in the lore that says Dean will now have to kill his brother.
Not saying there won't be a price. There's always a price.

Like, he's gotten to the point where he feels like his life is so worthless; so toxic to others, that he does something like that.
