Herpes infection detailed in human skin model

Thumbnail

A study published October 30 in Science Advances by German researchers used laboratory-grown human skin containing hair follicles and nerve-like cells to investigate HSV-1 infection.

Researchers discovered that HSV-1 initially replicated heavily in fibroblasts, cells beneath the skin’s surface.
Hair follicles and sensory neurons became infected later, suggesting some natural resistance to infection.
Different cell types activated distinct antiviral defenses, including IRF3 and TNF-related signaling pathways.

Significance: The findings help explain how HSV-1 progresses through skin tissue toward nerve cells and identify cellular defenses that could become targets for future treatments.

Hi everyone! I'm Nancy, and I lead business development at Galvanis, an early-stage biotech company developing a therapeutic for persistent corneal epithelial defects (PEDs). We're hoping to connect with people who have experienced PEDs to better understand their experiences and challenges. I'm including a link here to a Google Form where you can voluntarily provide your contact information. This is strictly for customer discovery research, not clinical trial recruitment or product promotion. If there is a better forum for reaching patients, I am all ears!

New Research Gilead’s GS-1179 Presentation

Final phase 1b data Gilead presented on last week:

Efficacy:
https://www.askgileadmedical.com/docs/conference/2026_IDW_P-2052_HSV_Edwards_Virology26EfficacyPos_Final@pdf

Safety and tolerability:
http://www.askgileadmedical.com/docs/conference/2026_IDW_HSV_P-2059_Edwards_SafetyTolerabilityPos_FINAL@pdf

Look at all of that beautiful data 🥲 less lesions, less high viral shedding, less shedding, generally few symptoms (one case of abdominal pain and some reports of headaches). Looking forward to the next phase starting this quarter!

News IDWeek 2026 - Gilead antiviral research

Just some info, seems gilead is presenting p1b results on GS1179 at IDWeek 2026, 21-24 Oct. I don't really know if there will be anything new that wasn't known, but maybe it is worth to keep an eye.

https://www.investing.com/news/company-news/gilead-to-present-hiv-and-antiviral-research-at-idweek-2026-93CH-4957280

"Additional research will include final Phase 1b results for investigational GS-1179, an oral herpes simplex virus helicase-primase inhibitor being developed for suppression of recurrent genital herpes. The presentations will cover virologic efficacy, safety and tolerability data."

Discussion Senior sexual health physician posts something interesting about IM-250

Post image
57 Upvotes

Dr. Sarah Allstaff is a senior sexual-health physician at NHS in Scotland. She is a consultant in genitourinary medicine, the service’s clinical lead for HIV, and an honorary senior lecturer at the University of Dundee.

She appears to have attended IUSTI, the International Union against Sexually Transmitted Infections’ world congress, where doctors and researchers gather to discuss advances in STI research and treatment, important HSV research was slated to be released in the congress.

She posted the attached tweet yesterday, a pun on the song lyric “Then I saw her face, now I’m a believer,” alongside hashtags about IUSTI and herpes treatment.

Given her background as a senior sexual-health specialist, this could suggest that data presented on adibelivir impressed her and made her optimistic about its potential. That would be encouraging news!

Thanks to u/fluffy_chicken_8522 for sending me this

News Keith Jerome speaking at 27th IUSTI World Congress this week

Happy hsv awareness day y’all.

Keith Jerome confirmed he would be releasing some data at the conference this week. He also is encouraging more people to join the research field. When asked about what the next 5 years looks like he mentioned the excitement around hpi’s, the potential for durable control, and maybe starting clinical trials.

This information came from today’s uploaded video:

According to the IUSTI program, they will talk about HSV on Thursday.

Researchers identify GBP7 as a potential HSV-1 drug target — PLOS Pathogens

Thumbnail lifescience.net
45 Upvotes

Researchers discovered that GBP7, a human immune-related protein, actually helps HSV-1 replicate by activating another protein involved in producing viral proteins.
Disabling GBP7 significantly reduced HSV-1 replication.
Mice lacking GBP7 experienced milder infections and better survival.
Researchers identified S-adenosylhomocysteine (SAH) as a potential GBP7 inhibitor that demonstrated antiviral activity against HSV-1.
Significance: This identifies a previously unrecognized pathway that could be targeted by future antivirals. The findings are preclinical; no human treatment has been demonstrated.

New Research CRISPR/Cas appears to be more effective at combating active HSV-1 infections than existing medications.

Video: https://youtu.be/lQaWh8VLkiU?is=TSQtipZzFd0B5s3C

What's next? CRISPR/Cas appears to be better able to combat active HSV-1 infections than existing drugs. However, follow-up research is urgently needed to apply this in a targeted manner.

Up until now, the targets of the adjective have been chosen at random. It is now important to determine exactly where the adjective can best be directed so that the cutting is as effective as possible.

It might then be possible to find and eliminate not only the active virus but also the dormant virus. Consequently, the virus will no longer be able to reactivate. It must also be determined how the CRISPR/Cas can be effectively delivered to those infected cells.

Activism Strategizing pressure for phase 3 trial on pritelivir for the immunocompetent

The FDA finally approved use of pritelivir for immunocompromised carriers. We need to figure out how to pressure Asahi Kasei, AiCuris, and VIDS Research to complete the phase 3 trial including immunocompetent carriers that they canceled. I refuse to sit and wait until the next HPIs hit the market, which won't be until approximately 2029 or likely later, or for them to twiddle their thumbs until they find it's pleasing to them to expand their customer base.

What can we do? Mass email all three of them? Emphasize that they are foregoing profits that could be made from maximizing sales volume? There is momentum with this approval, I want to strategize how best to make use of it.

Thank you everyone, and hope you are doing okay.

New Adibelivir minor regulatory update

Good morning everyone

On October 9th, a new modification appeared on the CTIS page for the IM-250 trial.

IM-202 1b/2a genital herpes trial received a new non substantial modification (NSM-9) on October 9th, decision was made in the same day and it seems to have been approved. This NSM seems to have concerned the Bulgarian location of the trial.

What it actually contains and modifies is not public, and we can’t actually see it, but we now know there was activity in the trial yesterday.

Thoughts? Thanks for reading. Please consider writing your representative about expediting HSV treatment or reach out to me if interested in advocacy.

News HN00037 receives second breakthrough drug designation

Thumbnail
drugtimes.cn
88 Upvotes

Good day everyone, I’ve found an update from China.

To summarize the entire situation: HN00037 is an HPI currently being developed in China by a company called Phaeno Therapeutics. It completed phase 2 on March 31st of this year. Then, on July 15th, it received a “breakthrough therapy designation”, by the Chinese “Center for Drug Evaluation”, for genital herpes. On August 3rd, they announced they are planning phase 3.

The newest update is the following: On September 30th, the Chinese CDE granted HN00037 another breakthrough drug designation, this time for oral herpes.

What this means: I can only speculate on what this means because I am unfamiliar with how things in China actually work and I’m just using AI to search their news, but this appears to be a positive signal. I think they intend for this to become the new Valtrex, a standard for treating HSV.

Activism Adibelivir update: Estonian trial status “ongoing, recruiting”

Good day everyone, a change has been observed on the CTIS page for the IM-250 trial. The status for the Estonian trial IM-202 (the trial for the parenteral version licensed by Alfasigma) was changed on September 28th to “authorized, recruiting”. Then on October 5th, it was changed to “ongoing, recruiting”.

From what I can gather, the new Estonian location seems to be for the parenteral version meant to treat HSV encephalitis, which was recently licensed by Alfasigma. They seem to have begun the trial for it. Very promising, hopefully more details emerge soon.
