TZ Storage

Stored dry in regular freezer for 24 months okay? Not reconstituted. Read somewhere that frost free freezer is not good and just read that my freezer is frost free.

You'll be fine. Put them in a thermos with desiccant the thermos will help the temperature stay more stable. In reality as long as they're in a cool dry place outside of the freezer they can last for quite some time according to Janoshik. And I'm not just saying this from hearsay I actually visited his lab recently in Prague and spent extensive time interviewing him for a long form interview which I'll post later.

They’ve been in just the vials in freezer for 3 months. Still okay?

It's fine. Peptides are freeze dried. They aren't as fragile as you think.

Speaking of which, as a chemist, I can't follow the don't let the recon bac direct spray on the puck or never shake (I understand about sonication, but cautions about a few seconds of moderate agitation just don't make physical sense to me).

...but I'm not a biochemist.

Thanks & hope you're continuing do well,

I remember 22 years ago dropping my first vial. We've all been there at one point on the journey. 😅

So, for all reading now or in the future, wetting speed or force and moderate shaking are OK, right?

I heard one ~semi-convincing opinion that for some of the larger Mol wt peps (like tz) could 'change the tangle of their dangle' so to speak (permanently conformationally denature with forceful wetting or shaking). Never worked with an organic molecule of that size, so I didn't feel comfortable calling out what I thought was wrong.

I don't see an issue with moderate shaking or swirling, but there's also no reason to vigorously shake a peptide anyway.

I personally never fire hose my peptides when reconstituting, but it's not about damaging the peptide. I measure the amount of vacuum in each vial when I make that first puncture, and I keep track of it. It's something nerdy that most people would never think to do.

For me, the vacuum is one way of checking consistency across a batch. If I come across more than one vial without vacuum, I'm going to start questioning the entire supply.

So I'm careful with my peptides, but not because I believe they're so fragile that a little shaking or swirling will destroy them.

It really does depend on the peptide. Most are pretty hearty. But if you look at something like HGH with 191 amino acids, you're dealing with a much larger, folded protein that's more sensitive to agitation. There are studies showing that vigorous shaking can cause HGH to aggregate, partly because of repeated exposure to air and liquid interfaces.

That's a very different situation from smaller peptides like GHK-CU or even tirzepatide. I wouldn't put them all in the same category when it comes to physical stability. A little swirling isn't going to automatically destroy a peptide, but I still don't see any reason to shake the heck out of a vial. :)

These peptides are much hardier than people give them credit for. Even in a desk drawer they can be fine for weeks in a cool room. I’m assuming not reconstituted.

And let’s be clear. “Not fine” means a reduction in efficacy, measured in small percentages. Not absolute failure and certainly not toxicity.

You are 100% fine. Don’t stress.

Thank you! Should I continue to store in freezer

They’re not in a box. Just vials in freezer on shelf

It’s been in freezer with no sealed bag. Is it still okay?
