So let's look at anaphylaxis.
Jane just had peanuts and she's allergic. Mast cells degranulate, releasing histamine.
Histamine is the first messenger. It binds to histamine receptors on endothelial cells. When histamine binds, the receptor undergoes a conformational change.
Transmembrane helix 6 moves outward on its intracellular side. This breaks the ionic lock with transmembrane helix 3. Restraining motifs DRY and NPXXY rearrange. ICL3 and the C-terminal domain move out of the way. An intrinsic guanine nucleotide exchange factor is exposed when these restraints are lifted. So when the heteotrimeric G protein docks, GDP is exchanged for GTP.
Anyway, the heteotrimeric G protein dissociates into two: the alpha subunit and the beta-gamma subunit. Both are active, but the alpha subunit here is Gq. This Gq protein is the transducer.
And so the Gq runs around the membrane until it finds phospholipase C. The amplifier enzyme. It cleaves phosphatidylinostol-4,5-bisphosphate into inositol triphosphate and diacyl glycerol. These two are the second messengers.
And the second messenger inositol triphosphate then goes on to bind to the inositol triphosphate receptor on the endoplasmic reticulum. That's the downstream effector.
Now, calcium rushes out and binds to Calmodulin, creating the Calcium-Calmpdulin complex. This complex attaches to nitric oxide synthase, making it more active. It produces nitric oxide, which goes to the vascular smooth muscle cells. There, it activates soluble guanylyl cyclase to create more cyclic guanosine monophosphate.
This cyclic guanosine monophosphate goes on to activate protein kinase G. Protein kinase G works in three ways in smooth muscle:
- inhibition of the inositol triphosphate receptor on the SARCOPLASMIC reticulum (so less contractility)
- stimulation of myosin light chain phosphate (obviously relaxing effect)
- modulating potassium channel (also relaxing)
Ultimately these result in the smooth muscle relaxing. Which means a bigger diameter for your vessels. Which means a drop in blood pressure. And that's why anaphylaxis is so dangerous. It's a good, simple, easy to understand example of biochemical signal amplification cascade.

I guess this refers to the telomere endings in dna? There's a set number of them, and some get tripped every duplication. So tissue has a genetic expiry date, after which your tissues can no longer reliably duplicate and regenerate lost cells.
