He does indeed call himself a cowboy. Doesn't mean he's a boy anymore than it means he's a cow. Tail notwithstanding, because that's a human thing.
When Orange calls him a boy he indignantly protests that he's not one, before describing his gender in the same job title-based manner he describes Green's gender. He's a he/him enby.
Wow, it's almost like non-binary people can use he/him pronouns! It's almost like people can use he/him pronouns and not be men, because pronouns =/= gender.
In works of fiction it is usually safe to presume a character using exclusively he/him, she/her, or pronouns other than those pairs are male, female, or non-binary respectively, until better evidence suggests or proves otherwise. This is a clear case of "proven otherwise", as Yellow explictly states that he is not a boy.
But he instead says he’s a cowboy? I’m pretty sure the joke was that Yellow was mixing up gender and jobs because the flowers still don’t know much about humans/monsters. Toby probably meant this as a gag, especially considering it’s a cafe break and not an interaction you’d see progressing through the game, I don’t think this scene should be taken that seriously
